Bradykinin production associated with oxygenation of the fetal lamb.

چکیده
Previous studies showed that synthetic bradykinin produced in-vitro constriction of the ductus arteriosus of the fetal lamb and guinea pig, constriction of isolated human and lamb umbilical vessels, and pulmonary vasodilatation in the fetal lamb. The present study showed that bradykinin precursor, kininogen, is present in arterial blood of the fetal lamb by 61 days of gestation and that its concentration increases toward term. We studied the bradykinin-generating system in 6 exteriorized fetal lambs. Kininogen concentrations in left atrial blood decreased within 1 to 2 minutes after beginning ventilation with O2, and free bradykinin was detected in left atrial blood. Pulmonary arterial blood kininogen concentration was not significantly altered. Kininogen concentration in left atrial blood did not fall in 4 other lambs ventilated with No but did fall after subsequent ventilation with O2. To study the effects of increased oxygenation in utero without lung expansion, 6 pregnant ewes were exposed to hyperbaric O2 at 3.63 atmospheres absolute. Associated with a mean increase in brachial arterial blood Po2 to 44 mm Hg, a value equivalent to that in the ventilated exteriorized fetus, kininogen concentration fell, and free bradykinin was detected in the brachial arterial blood of 3 of the 6 fetuses. In 4 other exteriorized lambs we measured kininogen concentration in brachial arterial and right ventricular blood at frequent intervals up to 30 minutes after ventilation with O2. The concentration in brachial arterial blood fell before that in right ventricular blood. The hypothesis is presented that bradykinin is produced from kininogen in the lungs of the fetal lamb when oxygenated, and that the maximal rate of production occurs during the first few minutes after expansion of the lungs or exposure of the ewe and fetus to hyperbaric O2. The Role of Mas Receptor and Bradykinin on Nitric Oxide Production Response to Angiotensin 1-7 in Ovariectomized Rats Treated with Estradiol
Introduction: The angiotensin 1-7 (Ang1-7) Mas receptor (MasR) axis, bradykinin (BK) and female sex hormone are involved in releasing of vasodilatory biomarkers including Nitric Oxide (NO). We examined the role of MasR and BK on NO metabolite (nitrite) production response to Ang 1-7 infusion in ovariectomized rats treated with estradiol. Methods: A total of 48 female Wistar rats were divided i...
متن کاملPreparation of a Photoactivable Ligand for Bradykinin Receptors
ABSTRACT Most of physiological effects of bradykinin is due to its effect on bradykinin B2 receptors. There is no reports of preparation of a photoactivatable analogue of bradykinin reactive with sloubilized bradykinin B2 recpetors. Photoactivatable radioactive ligands are powerful tools in purification of receptors. In this study we report preparation of a photoactivatable iodinatable analo...
متن کاملA case presentation of spider lamb syndrome in a Kermanian breed lamb
Skeletal abnormalities are most often used to describe defects in the arms or legs that are associated with genes or chromosomes, or that occur due to an event that happens during pregnancy. Spider lamb syndrome (SLS) is a congenital disorder in sheep breeding that is recognized by some deformities in skeletal system especially in the limbs.A dead day-old cross-breed white lamb with deformed li...
متن کاملCongenital lumbar vertebrae agenesis in a lamb
Congenital agenesis of lumbar vertebrae was diagnosed in a day-old female lamb based on radiology and clinical examinations. There was no neurological deficit in hindlimb and forelimb associated with standing disability. Radiography of the abdominal region revealed absence of lumbar vertebrae. Necropsy confirmed clinical and radiographic results. No other anomaly or agenesis was seen macroscopi...
متن کاملMaternal and Fetal Acid-Base Chemistry: A Major Determinant of Perinatal Outcome
Very small changes in pH may significantly affect the function of various fetal organ systems, such as the central nervous system, and the cardiovascular system with associated fetal distress and poor Apgar score. Review of existing data on maternal-fetal acid-base balance in pregnancy highlight the factors that are associated with derangements of the acid-base status and the impact of the dera...
